Subtractive Immunization as a Method to Develop Respiratory Syncytial Virus (RSV)—Specific Monoclonal Antibodies

نویسندگان

چکیده

Respiratory Syncytial Virus (RSV) is a significant cause of lower respiratory tract infections in the young, elderly, and immunodeficient patients. As such, virus represents an important morbidity mortality worldwide. Development monoclonal antibodies against RSV has resulted commercial prophylaxis, palivizumab (Synagis®), different that have improved our understanding structure viral proteins. In this study, immunization technique, subtractive immunization, was evaluated for its applicability to develop RSV-specific antibodies. One hybridoma which produced with strongest staining infected cells, ATAC-0025, selected further characterization. This antibody belongs IgG1 class, neutralizing capacity recognizes envelope F-protein. The broad reactivity range reference strains clinical isolates.

Subtractive immunization yields monoclonal antibodies that specifically inhibit metastasis

Subtractive immunization allowed the isolation and characterization of monoclonal antibodies that specifically inhibit metastasis but not proliferation of highly metastatic human tumor cells. The tolerizing agent cyclophosphamide was used to suppress the immune system in mice to dominant immunodeterminants present on a non-metastatic variant (M-) of the human epidermoid carcinoma cell line (HEp...

Monoclonal antibodies to human respiratory syncytial virus and their use in comparison of different virus isolates.

Monoclonal antibodies to the RSN -2 isolate of human respiratory syncytial (RS) virus have been characterized with regard to their specificity for viral polypeptides and for different RS virus isolates. Five hybridoma antibodies recognized the phospho-protein VPP32 and the other recognized the matrix protein VPM27 . Evidence was obtained to support the view that VPP32 was associated with the nu...

Monoclonal antibodies to respiratory syncytial virus proteins: identification of the fusion protein.

Six monoclonal antibodies directed against respiratory syncytial virus proteins were produced. Each was characterized by immunoprecipitation and indirect immunofluorescence. One was directed against the nucleocapsid protein. NP 44, two were directed against a 37,000-dalton protein, two were directed against the major envelope glycoprotein, GP 90, and one was directed against the 70,000-dalton e...

Monoclonal antibodies to the fusion protein of bovine respiratory syncytial virus.

Five monoclonal antibodies specific for bovine respiratory syncytial virus were characterized by Western immunoblotting, radioimmunoprecipitation, and epitope mapping assays. The monoclonal antibodies were found to be specific for the fusion protein, and there were at least two antigen binding sites, one of which was neutralizing.

A Rapid Subtractive Immunization Method to Prepare Discriminatory Monoclonal Antibodies for Food E. coli O157:H7 Contamination

To detect food E. coli O157:H7 contamination rapidly and accurately, it is essential to prepare high specific monoclonal antibodies (mAbs) against the pathogen. Cyclophosphamide (Cy)-mediated subtractive immunization strategy was performed in mice to generate mAbs that react with E. coli O157:H7, but not with other affiliated bacteria. Specificity of 19 mAbs was evaluated by ELISA and/or dot-im...
